Alleviation of Adverse Effects of Copper on Allium cepa L. by Exogenous Ascorbic Acid Application

Abstract
In this study ameliorative effects of ascorbic acid (0.5 mM) on onion (Allium cepa L. cv. Red Azarshahr) under copper stress (Cu: 6.5 μM) were studied. Fresh weights of leaves and roots were reduced in seven week old plants under excess copper condition. Cu stress also reduced membrane stability index in leaves. There was a considerable increase in H2O2 content and lipid peroxidation in both roots and leaves of plants under excess Cu. Leaves anthocyanin contents, leaves and roots phenolic compounds, soluble proteins and proline contents were increased in response to Cu toxicity. The fresh weights of leaves and roots were improved during addition of ascorbic acid to root medium in stressed and non-stressed plants. Exogenous ascorbic acid significantly increased membrane stability index in Cu stressed plants. We find considerable reduction in anthocyanins, phenolic compounds and proline with ascorbic acid treatments in all plants. Ascorbic acid application also reduced H2O2 content and prevented lipid peroxidation in leaves and roots during presence or absence of excess Cu. Treatment of plants with ascorbic acid increased soluble proteins content only in non-stressed plants. The results demonstrate that root applied ascorbic acid ameliorated the copper induced oxidative adverse effects on onion growth.

در این پژوهش اثرات بهبودی اسید آسکوربیک 5/0 میلی مولار بر رقم قرمز آذرشهر پیاز خوراکی (Allium cepa L.) تحت تنش مس (5/6 میکرومولار) مورد مطالعه قرار گرفت. در گیاهان یک هفته­ای پیاز تیمار مس موجب کاهش وزن تر ریشه­ها و برگ­ها گردید. تنش مس منجر به کاهش شاخص پایداری غشاهای سلولی برگ­ها و افزایش قابل ملاحظه در محتوای پراکسید هیدرو‍‍ژن و پراکسیداسیون چربی­های ریشه­ها و برگ­ها شد. در پاسخ به سمیت مس محتوای آنتوسیانین برگ­ها، ترکیبات فنلی برگ­ها و ریشه­ها، ونیز مقدار پروتیئن­های محلول و پرولین افزایش نشان دادند. تیمار با اسید آسکوربیک موجب افزایش وزن تر ریشه­ها و برگ­ها در همه گیاهان در شرایط واجد تنش و نیز فاقد تنش گردید. در گیاهان تحت تنش مس، کاربرد برون زای اسید آسکوربیک به طور معنی داری شاخص پایداری غشاهای سلولی برگ­ها را افزایش داد. در همه گیاهان تیمار اسید آسکوربیک کاهش قابل ملاحظه آنتوسیانین­ها، ترکیبات فنلی و پرولین را باعث شد. کاربرد آسکوربیک اسید موجب کاهش محتوای پراکسید هیدرو‍‍ژن و ممانعت از پراکسیداسیون چربی­ها در ریشه و برگ همه گیاهان در هر دو شرایط واجد و فاقد تنش شد. در گیاهانی که تحت تنش مس قرار نداشتند تیمار با اسید آسکوربیک منجر به افزایش محتوای پروتیئن­های محلول شد. نتایج حاکی از آن است که کاربرد ریشه­ای اسید آسکوربیک اثرات مضر اکسیداتیو مس بر رشد پیاز خوراکی را تقلیل می­دهد.
